三、HTRF檢測(cè)應(yīng)用中多功能熒光酶標(biāo)儀的選型要略

        常規(guī)FI、TRF、FRET檢測(cè),人們可能只關(guān)注到熒光酶標(biāo)儀的激發(fā)、發(fā)射波長(zhǎng)配置與測(cè)試要求契合的問題。而當(dāng)了解HTRF標(biāo)準(zhǔn)流程后,對(duì)優(yōu)化熒光檢測(cè)條件的意義和其中涉及的技術(shù)要素之復(fù)雜,興許會(huì)有感悟。

       開展HTRF測(cè)試前,實(shí)驗(yàn)者首先須確認(rèn)是否有可利用的HTRF認(rèn)證多功能熒光酶標(biāo)儀資源。

       確認(rèn)多功能熒光酶標(biāo)儀屬HTRF認(rèn)證型號(hào)后,還應(yīng)核實(shí)是否配置有HRTF檢測(cè)規(guī)定的專用光學(xué)元件。不同品牌機(jī)型,制造商提供的HTRF功能組件有區(qū)別。

        如VantaStar、Folarstar OmegaSpectraMax iD5、Synergy NEO2均須添加HTRF專用的具有Z軸調(diào)節(jié)功能的增強(qiáng)型時(shí)間分辨熒光模塊(Enhanced TRF module)、320/340nm紫外激發(fā)濾片、520/620/665nm發(fā)射熒光濾鏡這三件套。

       相對(duì)而言,MD SpectraMax i3、SpectraMax ParadigmSpectraMax i3x三款熒光酶標(biāo)儀,由于采用的是檢測(cè)功能模塊化設(shè)計(jì),必要時(shí)直接將選配的HTRF檢測(cè)卡盒插入主機(jī)卡槽就可以啟動(dòng)測(cè)試。實(shí)驗(yàn)者無(wú)須為激發(fā)與發(fā)射濾光片的波長(zhǎng)與帶寬、二向分色鏡截止波長(zhǎng)范圍這類問題糾結(jié)。

四、HTRF檢測(cè)程序與同類TR-FRET方法間的兼容性

       目前,同樣是基于TR-FRET技術(shù)開發(fā)的檢測(cè)試劑, HTRF之外還有MD IMAP Assays、BellBrook Transcreener assay 、Invitrogen LanthaScreen TR-FRET Kinase Assay、Bioauxilium Thunder Enhanced TR-FRET assay、PE Lance TR-FRET Assays等。

       單論測(cè)試光譜數(shù)據(jù),這些TR-FRET試劑的檢測(cè)光譜設(shè)置基本是通用的。那實(shí)際工作中,其他同類產(chǎn)品能否用HTRF的檢測(cè)標(biāo)準(zhǔn)程序測(cè)定?

       分析結(jié)果是:不行。

       PE-Cisbio強(qiáng)調(diào)的是,HTRF認(rèn)證測(cè)試流程、方法的應(yīng)用范圍嚴(yán)格限于HTRF試劑和技術(shù)使用,不適用于用于其他TR-FRET產(chǎn)品的測(cè)定。

       TRF-FRET檢測(cè)具有時(shí)間高度靈敏的特性。在各種不同來(lái)源樣品測(cè)試條件下,不同TRF-FRET試劑盒的熒光光譜穩(wěn)定性、半衰期特征的差異,注定了檢測(cè)過程的積分時(shí)間、脈沖數(shù)量、時(shí)間延遲幾個(gè)關(guān)鍵參數(shù)難以完全匹配。盡管兩種試劑盒測(cè)定的激發(fā)波長(zhǎng)、發(fā)射波長(zhǎng)峰值特征一致,但不代表TRF-FRET過程中供體、受體兩種熒光信號(hào)的動(dòng)力曲線相同。

       HTRF試劑采用有大環(huán)結(jié)構(gòu)的穴狀態(tài)鑭系配合物,其光譜穩(wěn)定性優(yōu)異,與特殊優(yōu)化的緩沖液配合,可以實(shí)現(xiàn)0.6-1ms的超長(zhǎng)熒光壽命。其他的TR-FRET技術(shù)中所用的鑭系配合物、FRET能量受體釋放的FRET熒光信號(hào)動(dòng)力學(xué)特征除非能高度還原HTRF產(chǎn)品FRET過程的熒光表現(xiàn)。否則,在同一型號(hào)熒光酶標(biāo)儀上用HTRF提供的標(biāo)準(zhǔn)測(cè)試程序測(cè)定其他廠商試劑,難免弄巧成拙、東施效顰。

      對(duì)比Ciosbio HTRF-Eu/red Assay、LanthaScreen Eu Kinase Activity Assay在Perkin Elmer EnVision多功能熒光酶標(biāo)儀上檢測(cè)所用的測(cè)試參數(shù),可發(fā)現(xiàn):盡管兩個(gè)試劑盒規(guī)定的激發(fā)波長(zhǎng)、雙熒光發(fā)射波長(zhǎng)設(shè)置基本一致(測(cè)試年份不同,同一型號(hào)儀器出廠預(yù)裝濾光片規(guī)格略有變動(dòng)),但脈沖信號(hào)的積分時(shí)間延遲(HTRF-60μs VS LanthaScreen-100μs)、積分時(shí)間長(zhǎng)度(HTRF-400μs VS LanthaScreen-200μs)的設(shè)定迥異。

        這證實(shí)了我們分析的結(jié)論:不同TR-FRET試劑盒在熒光多功能酶標(biāo)儀上的規(guī)定的測(cè)試條件設(shè)置存在較大區(qū)別,其標(biāo)準(zhǔn)檢測(cè)程序相互難以有效兼容。
        表5的信息還表明:與Ciosbio HTRF-Eu比,LanthaScreen Europium激發(fā)的背景熒光衰減慢、而FRET供體和受體的熒光壽命短;LanthaScreen Eu檢測(cè)時(shí)間延遲拉長(zhǎng),極大地限制了檢測(cè)積分窗口的選擇余地和測(cè)試靈敏度的提升空間。
